Holmes and Watson are visited by Dr. Mortimer, who consults them regarding the death of his patient, Sir Charles Baskerville, who believed an ancient curse on the family was being fulfilled. Mortimer is in London to accompany the Baskerville heir Henry back to his new home, but sinister forces are at work, including a warning to stay away, and a mysterious theft at Sir Henry's hotel.

Dr. Mortimer and Sir Henry ask Holmes to return to Devonshire with them to unravel the mystery behind Sir Charles' death, but Holmes claims previous commitments and sends Watson in his place. At Baskerville Hall the butler and his wife are acting strangely. While on the moor Watson meets Mr. Stapleton out with his butterfly net, as well as his sister who mistakes him for Sir Henry and implores him to leave the area. Another stranger is seen lurking on the moor and he might possibly be Seldon, the recently escaped convict from a nearby prison.

Two mysteries are solved when Sir Henry and Watson confront the Barrymores about their nocturnal activities and another neighbor (Franklin) visits and gives Watson a clue to the location of the mysterious man on the moor. A strange encounter with the Stapletons puts Sir Henry in an odd situation; and Holmes finally makes his way to Baskerville Hall when another death that might be attributed to the spectral hound occurs.

Holmes puts together disparate clues to the identity of the person behind the deaths and brings Inspector Lestrade from London to aid in the capture. Holmes puts Sir Henry in mortal danger in order to spring a trap, but it results in yet another death as the culprit is unmasked and his scheme exposed.
